Research by scientists at Pennsylvania State University, Stanford University and an international team of collaborators found that blocking an enzyme called indoleamine-2,3-dioxygenase 1 (IDO1) restored brain astrocyte metabolism in a mouse model of AD and memory and brain function were rescued.
The results suggest that IDO1 inhibitors are currently being developed as treatments for a variety of cancers – including melanoma, leukemia and breast cancer – as treatments for early-stage AD and potentially other neurodegenerative diseases. They could be used for the first time as treatments for chronic diseases.

Inhibiting this enzyme, especially with compounds that have already been tested in human cancer clinical trials, would be a major step in finding ways to protect our brains from the damage caused by aging and neurodegeneration. “We show that there is great potential for IDO1 inhibitors, which are already in development for the treatment of cancer,” Katrin Andreasen, MD, the Edward F. and Irene Pimley Professor of Neurology and Neurological Sciences at Stanford University School of Medicine, told the Drug Directory.
